Pharmacological properties
NexGard Spectra refers to combined antiparasitic drugs.
The active ingredients of the drug – afoxolaner and milbemycin oxime provide its wide range of antiparasitic action against ectoparasites and nematodes parasitizing in dogs: fleas (Ctenocephalides felis, Ctenocephalides can is); ticks (Dermacentor variabilis, Dermacentor reticulatus, Rhipicephalus sanguineus, Ixodes scapularis, Ixodes ricinus, Ixodes holocyclus, Haemaphysalis longicornis, Amblyomma americanum); gastrointestinal nematodes (Toxocara canis, Toxascaris leonina, Ancylostoma caninum, Ancylostoma braziliense, Trichuris vulpis); larvae of dirofilariae (Dirofilaria immitis).
Afoxolaner is an insectoacaricide of the isoxazoline group, the mechanism of its action is to block GABA-dependent receptors in arthropods, hyperexcitation of neurons, impaired transmission of nerve impulses, which leads to paralysis and death of ectoparasites. After oral administration, afoxolaner is easily absorbed in the gastrointestinal tract and reaches the systemic circulation, the maximum plasma concentration is observed 2-4 hours after administration, the bioavailability is 88%. Afoxolaner is slowly metabolized with the formation of hydroxylated compounds, excreted from the body mainly in the bile and slightly in the urine. The half-life of afoxolaner is 14 days.
Milbemycin oxime is a macrocyclic lactone, active against larvae and adults of nematodes parasitizing in the gastrointestinal tract of dogs, as well as larvae of Dirofilaria immitis. The mechanism of action of milbemycin is due to an increase in the permeability of cell membranes for chlorine ions (SG), which leads to overpolarization of the membranes of the cells of the nervous and muscle tissue, paralysis and death of the parasite. The maximum concentration of milbemycin oxime in the blood plasma of dogs is reached within 1-2 hours, the bioavailability is 81%. The compound is excreted from the body mainly unchanged, the half-life is about 1.5 days.
The insecticidal effect of the drug begins 30 minutes after its oral administration, the death of fleas and ixodid ticks on the animal occurs, respectively, for 6 hours and 24 hours. A single use of the drug prevents re-infestation within 1 month; the death of fleas occurs before oviposition, which prevents contamination of the premises with eggs and insect larvae.
NexGard Spectra, according to the degree of impact on the body, belongs to low-hazard substances (hazard class 4 according to GOST 12.1.007-76), in recommended doses it does not have teratogenic, embryotoxic, mutagenic properties, does not have a local irritating effect. The drug is toxic to bees, as well as fish and other aquatic organisms.
