The composition of the drug
The drug in the form of lyophilized powder for injection of white or light yellow color complete with solvent (water for injection) is available in glass vials sealed with chlorobutanol caps and wrapped in aluminum caps.
Each vial contains an active substance: 250 mg tiletamine hydrochloride and 250 mg zolazepam hydrochloride. Excipients – sodium sulfate and lactose.
Pharmacological properties
Tiletamine is a general dissociative anesthetic that causes a pronounced analgesic effect, but insufficient muscle relaxation.Β Tiletamine does not suppress pharyngeal, laryngeal, cough reflexes, does not suppress the respiratory system.
Zolazepam suppresses the subcortical areas of the brain, causing anxiolytic and sedative effects, relaxes the striated muscles.Β Zolazepam enhances the anesthetic effect of tiletamine.Β It also prevents cramps caused by tiletamine, improves muscle relaxation and speeds up recovery after anesthesia.
The time of anesthesia after intramuscular administration is individual, so after injection of the drug should be established careful monitoring of the animal.
In cats, the onset of action of the drug after intramuscular administration is observed in 1-7 minutesΒ The duration of anesthesia depends on the dose, but the maximum effect is usually observed after 20-60 minutes.Β The recovery period after anesthesia varies from 1 to 5.5 hours.
In dogs, the onset of action of the drug after intramuscular administration is observed in an average of 7.5 minutesΒ The average duration of anesthesia is about 30 minutes, the recovery period is up to 4 hours.
In case of overdose, as well as in very young and old animals, the recovery period is longer.Β The active substances of Zoletil are metabolized in the liver and excreted in the urine.
Indications for use
General anesthesia for diagnostic procedures and surgery in cats and dogs.

The antiparasitic collar should be removed from the animal 24 hours before anesthesia and a 12-hour fasting diet should be maintained.
15 minutes before administration of the drug, it is recommended to subcutaneously administer atropine sulfate in a dose: dogs – 0.1 mg / kg body weight, cats – 0.05 mg / kg body weight.
Dilute the contents of the powder vial with solvent.Β At intramuscular administration loss of rectifying reflexes occurs in 3-6 min., At intravenous administration – in 1 min.
Intramuscular injection:
Dogs: clinical examination – 7-10 mg / kg;Β short-term general anesthesia for minor surgery – 10-15 mg / kg;Β long general anesthesia for large and painful surgeries – 15-25mg / kg.
Cats: clinical examination – 10 mg / kg;Β long-term general anesthesia for large and painful surgeries – 15 mg / kg.
Intravenous administration:
Dogs: clinical examination – 5 mg / kg;Β short-term general anesthesia for minor surgery – 7.5 mg / kg;Β long-term general anesthesia for large and painful surgeries – 10 mg / kg.
Cats: clinical examination – 5 mg / kg;Β prolonged general anesthesia for large and painful surgeries – 7.5 mg / kg.
If necessary, zoletil can be re-introduced in a dose not exceeding 1 / 3-1 / 2 of the initial dose.
Zoletil has no cumulative effect and can be injected repeatedly, in doses not exceeding 1 / 3-1 / 2 of the initial dose.Β The total dose of the drug should not exceed the safety threshold: 30 mg / kg for dogs and 72 mg / kg for cats;Β in both species the minimum lethal dose is 100 mg / kg.Β The duration of anesthesia is from 20 to 60 minutes
Recovery from anesthesia: The analgesic effect of Zoletil is longer than that caused by surgical anesthesia.Β Exit from anesthesia is gradual (2-6 hours) and calm in the absence of noise and bright light.Β In cases of overdose, as well as in very young and old animals, the recovery period is longer.
Contraindication
Zoletil is contraindicated in animals with diseases of the pancreas, with severe disorders of the cardiovascular and respiratory systems, hypertension.Β In animals with kidney disease, the dose should be reduced.Β Zoletil penetrates the placenta and can cause respiratory depression in the fetus.
Side effect
Apnea may occur with high doses or intolerance to zoletil.Β During awakening, vomiting, excessive salivation and bronchial secretion (if premedication with atropine has not been performed), involuntary muscle twitching, hypertension, muscle rigidity may be observed.
In case of overdose, artificial lung ventilation is performed.Β Other symptoms are relieved by symptomatic therapy.
Special instructions
Concomitant use of zoletil and chloramphenicol prolongs anesthesia in cats for an average of 30 minutes.
Concomitant use of zoletil with phenothiazine (acepromazine) causes respiratory and cardiac depression.Β At simultaneous use of zoletil with barbiturates or inhalation anesthetics the dose of the last should be reduced.
Storage conditions
List B. 2 years from the date of manufacture at a temperature of +5 to + 20 Β° C. The solution is stored for 8 days in a dark place at a temperature of + 4 Β° C, at room temperature – 24 hours.
